DPPH, a stable free radical, is a dark purple prismatic crystal. The maximum absorption wavelength is 528 nm.
DPPH has three different crystalline forms, their lattice symmetry and melting point (m.p.) There are differences. The results are as follows: DPPH- Ⅰ (m.p. 106 °C) is an orthorhombic system, DPPH- Ⅱ (m.p. 137 °C) is amorphous, DPPH- Ⅲ (m.p. 128-129 °C) is a triclinic system. The common commercial powders are mixtures of different crystalline phases with melting points of 127-129 °C.
DPPH method was proposed for quantitative determination of antioxidant capacity of biological samples and foods in 1958. The DPPH radical has a single electron and its alcohol solution is purple and has a strong absorption at 517nm. In the presence of free radical scavengers, the absorption of free radical scavengers gradually disappears due to their single electron pairing, and the fading degree of free radical scavengers is quantitatively related to the number of electrons they receive, so a rapid quantitative analysis can be carried out by spectrophotometer.
